We currently have to access our DMAs through our corporate VPN. We believe that this is the cause of the DIS Injector failing (since our colleagues have been able to get it to work locally).
Are the any specific port configuration on developer or DMA (and any firewalls between) that needs to be set up for DIS to work?

I believe the default port is 8004, so if that's blocked, I could see that causing a problem for you. According to the help when setting up the DMA you can configure the HOST settings as follows:
Host
In this box, enter the IP address or server name of the DataMiner Agent using the following syntax:
https://[IP address or server name]:[Port]/SLNetService
Note:
•Both HTTP and HTTPS are supported.
•Specifying the IP port is optional. Default port: 8004
•Specifying the suffix “/SLNetService” is optional.
